Development of ketoprofen-p-aminobenzoic acid co-crystal: formulation, characterization, optimization, and evaluation

In the present study, ketoprofen-p-aminobenzoic acid (KP-PABA) co-crystal was prepared, to advance solubility and dissolution rate of drug, by solvent evaporation technique employing central composite experimental design. The optimized batch as recommended design characterized FTIR, DSC, XRD, SEM, NMR studies further, evaluated for in-vitro in-vivo anti-inflammatory analgesic activities. % drug release different batches found be between 34.20–60.11 µg/ml 68.11–93.45%, respectively. Physical characterization X-ray diffraction spectra differential scanning calorimetric confirmed crystallinity prepared co-crystal. IC50 value formulation pure observed 248.79 524.40 µg/ml, respectively, displaying that possesses more percentage protection against protein denaturation than ketoprofen. results activity carried out rat paw edema method revealed preparation provided a significant inhibition in volume contrast standard wistar rats.
